The ASFS method (Accurate Stochastic Fluorescence Spectroscopy) and its concept
The new proprietary ASFS method determines the binding behaviour of freely diffusing molecules in solution avoiding surface related artefacts such as unspecific binding and steric hindrance.
It guarantees straightforward sample preparation (simple mix and measure assay formats) and circumvents tedious immobilization processes. Unlike other solution based method, the ASFS Technology provides access to very low analyte concentrations ( <10-10 molar) and offers more flexibility in the choice of reaction partners (e.g. concerning molecule size) because of its capacity to precisely distinguish molecular species.
It makes uses of the fact that the individual molecular properties (e.g. size, diffusion, velocity, quenching and brightness) are reflected in different "fluorescence footprints".
It introduces the concept of the effective volume which is a molecule specific parameter. It characterizes fluorescent molecules and allows to identify and quantify them even in complex mixture with high statistical signifiance.
